چکیده

The exact role of the endocannabinoid system (ECS) during spermatogenesis has not been clarified. Scientists at the University of Rome "Tor Vergata" (Italy) and at the Endocannabinoid Research Group – Consiglio Nazionale delle Ricerche (Pozzuoli, Italy) have used purified germ cell fractions representative of all phases of spermatogenesis and primary cultures of spermatogonia, to demonstrate that male mouse germ cells possess an active and complete ECS [Grimaldi et al. Proc Natl Acad Sci USA 2009;106:11131-6]. They found that an autocrine endocannabinoid signalling might be operational during spermatogenesis, and that the ECS is modulated during meiosis, with the levels of the endocannabinoid 2-arachidonoylglycerol peaking before meiosis. In addition, activation of type-2 cannabinoid (CB2) receptors was found to promote meiotic progression of germ cells, by increasing the expression of early meiotic prophase genes.

Possible inhibitory roles of endogenous 2-arachidonoylglycerol during corticotropin-releasing factor-induced activation of central sympatho-adrenomedullary outflow in anesthetized rats.

We previously reported that intracerebroventricularly (i.c.v.) administered corticotropin-releasing factor (CRF) (0.5-3.0 nmol/animal) dose-dependently elevates plasma noradrenaline and adrenaline through brain phospholipase C-, diacylglycerol lipase- and prostanoids-mediated mechanisms in rats. Diacylglycerol produced by phospholipase C from phospholipids can be hydrolyzed by diacylglycerol li...

The endocannabinoid system and pain.

The therapeutic potential of cannabinoids has been the topic of extensive investigation following the discovery of cannabinoid receptors and their endogenous ligands. Cannabinoid receptors and their endogenous ligands are present at supraspinal, spinal and peripheral levels. Cannabinoids suppress behavioral responses to noxious stimulation and suppress nociceptive processing through activation ...

Neuronal protease-activated receptor 1 drives synaptic retrograde signaling mediated by the endocannabinoid 2-arachidonoylglycerol.

Protease-activated receptor 1 (PAR1) is a member of the G-protein coupled receptors that are proteolytically activated by serine proteases. Recent studies suggest a definite contribution of PAR1 to brain functions, including learning and memory. However, cellular mechanisms by which PAR1 activation influences neuronal activity are not well understood. Here we show that PAR1 activation drives re...

[Endogenous cannabinoid receptor ligands--anandamide and 2-arachidonoylglycerol].

Marijuana has been used as a traditional medicine and a pleasure-inducing drug for thousands of years around the world, especially in Asia. Delta(9)-tetrahydrocannabinol, major psychoactive component of marijuana, has been shown to interact with specific cannabinoid receptors, thereby eliciting a variety of pharmacological responses in experimental animals and human. In 1990, the gene encoding ...

The endocannabinoid signaling system: a potential target for next-generation therapeutics for alcoholism.

Research into the endocannabinoid signaling system has grown exponentially in recent years following the discovery of cannabinoid receptors (CB) and their endogenous ligands, such as anandamide (AEA) and 2-arachidonoylglycerol (2-AG). Important advances have been made in our understanding of the endocannabinoid signaling system in various aspects of alcoholism, including alcohol-seeking behavio...
